Paraoxonase 1 Activity Could Be A Potential Biomarker For Liver Damage

Abstract Variations of PON1 activity (PON1) were studied according to the type of liver injury, to assess the interest of this parameter in the diagnosis of liver diseases. The study was performed in population of patients with different liver damages. The liver diseases adopted during our investigation were grouped into different syndromes and pathologies. The classification by pathology, according to the clinical, biological and radiological context, made it possible to distinguish five types of hepatic impairment. The determination of the activity of PON1 was carried out on Konélab, and the assay the other enzymes as well as of the biochemical parameters were carried out on the Cobas 6000 system.The PON1 activity was lower in patients compared to control group (p<0.007). The risk to have PON1 activity decreased was doubled in patients suffering from liver diseases (OR=2.201; IC à 95%:1.074-4.512; p=0.030). This risk was more important in cirrhotic patients (OR=4.44, 95% CI:1.78 to 11.05, p=0.001). A positive correlation between PON1 and albumin (r=0.349, p<0.001), or HDL-c (r=0.370, p<0.001) was found in total population of patients. However, PON1 was inversely correlated with AST (r=- 0.338, p<0.001), PAL (r =-0.205, p=0.038) and total bilirubin (r=-0.293, p=0.003). PON1 was significantly lower in patient groups that have three perturbed lipid parameters (p=0.034). The PON1 activity could be a reliable biomarker of the degree of liver injury and could be integrated into the hepatic exploration tests, to better highlight the suffering liver and contribute to the diagnosis of liver damage.
