Effect of Artemisia Annua on oxidative stress and antioxidant gene expression in Nile tilapia after chlorpyrifos toxicity

Abstract In this study, the toxic effects of chlorpyrifos (CPF) on fish antioxidant enzyme activity in serum and transcriptomic levels of oxidative stress-related genes in liver tissue and histopathology of the liver tissue of Nile tilapia were examined. Additionally, the effects of Artemisia Annua (A. annua) , either alone or in combination with the CPF-contaminated water, were investigated. To accomplish this goal, 120 fish were randomly assigned to four distinct experimental groups. In the control group (Cont group), fish were fed a formulated diet and reared in normal water without toxicity. In the chlorpyrifos group (Chlo group), fish were fed a formulated diet and reared in water containing chlorpyrifos at a concentration of 15 µg L⁻¹. Artemisia Annua group (Art group), fish were fed a formulated diet enriched with A. annua at 5 g kg⁻¹ and reared in normal water without toxicity. Artemisia Annua+ chlorpyrifos group (Art+Chlo group): Fish were fed a formulated diet enriched with A. annua and reared in water containing chlorpyrifos. The experiment was done over 30 days. The results of the present investigation indicated that exposure to CPF-contaminated water significantly increased malondialdehyde (MDA) levels and decreased catalase (CAT) and superoxide dismutase (SOD) activity as compared with the control group. Furthermore, CPF exposure led to upregulation of genes associated with oxidative damage, including heat shock protein 70 ( Hsp70 ) and apoptosis-related cysteine peptidase caspase-3 ( Caspase-3) . Moreover, CPF disrupted antioxidant gene expression by upregulating the Kelch-like ECH-associated protein 1( Keap1 ) and downregulating the nuclear factor erythroid 2-related factor 2 ( Nrf2 ), CAT , and glutathione peroxidase ( GPx ). Liver histopathological examination of the chlorpyrifos-intoxicated group showing marked necrotic changes within the pancreatic tissue associated with leukocytic cell infiltration and marked vacuolation of the hepatocytes. However, A. annua supplementation mitigated the deleterious effects of CPF through increased activity of serum antioxidant enzymes and decreased levels of lipid peroxidation. Additionally, A. annua supplementation downregulated the genes associated with oxidative damage, including Hsp70 and Caspase-3 . Also, it improved antioxidant gene expression by downregulating Keap1 and upregulating Nrf2 , CAT , and GPx . Moreover, the hepatopancreas of the A. annua+ chlorpyrifos group shows normal hepatocytes and decreased necrotic changes within the pancreatic acini with infiltration of hemosiderin pigment.
